IACUC / LARC STANDARD PROCEDURES
PRODUCTION OF SUBCUTANEOUS TUMORS IN RATS AND MICE
Tumor Size: The maximal size will depend to some extent on tumor position. Important factors determining size limitation are protection of the animals' ability to move around the cage normally and to reach and consume food and water. Tumor size must not exceed 2.0 cm at largest diameter in mice and 4.0 cm in rats. Larger size tumors should be specifically justified in the IACUC protocol.
Tumor Position: Select a site that will not compromise the animal's mobility. Backs and flanks are preferred sites.
Monitoring Schedule: This will depend to some degree on growth rate of the tumor, but should not be less than:
- Three times weekly until a palpable tumor nodule is present (0.5-0.75 cm in diameter).
- Daily thereafter (including weekends and holidays).
- If tumor growth is rapid in the few days before termination, twice daily monitoring should be considered.
Variables to be Monitored: General appearance; ability to move normally around cage and reach food and water, ability to eat and drink: grooming habits: skin color (pallor); lethargy; skin over tumor (breakdown and/or necrosis normally require euthanasia); aggression (animals should be housed individually if aggressive behavior is shown).
